Comparative Analysis of Glucose, Fructose and Sucrose in Fruit Juice Using Quantitative NMR and HPLC Techniques

This study aimed to quantitatively determine glucose, fructose, and sucrose using quantitative nuclear magnetic resonance spectroscopy (qNMR) employing both external and internal standard approaches. The analytical performance of the qNMR methods was evaluated in terms of accuracy and precision and compared with that of high-performance liquid chromatography (HPLC) in fruit juice samples. The results, assessed using Bland-Altman analysis, demonstrated excellent agreement between qNMR and HPLC, with limits of agreement (±1.96SD) falling within acceptable ranges. In terms of precision, expressed as %RSD, the qNMR internal standard method exhibited the highest precision (%RSD 0.03 - 2.56%), followed by HPLC (%RSD 0.11 - 3.26%) and the qNMR external standard method (%RSD 0.29 - 2.77%), respectively. These findings confirm that qNMR and HPLC yield statistically consistent results. Moreover, the qNMR technique offers practical advantages, including simplified sample preparation, the elimination of calibration curve requirements, and significantly reduced analysis time compared to HPLC. Given its capability to simultaneously quantify multiple sugars in a single step, qNMR represents an efficient and convenient analytical tool for routine quality control of glucose, fructose, and sucrose in fruit juice products.
